    Time‐restricted feeding prevents depressive‐like and anxiety‐like behaviors in male rats exposed to an experimental model of shift‐work

    Individuals who regularly shift their sleep timing, like night and/or shift‐workers suffer from circadian desynchrony and are at risk of developing cardiometabolic diseases and cancer. Also, shift‐work is are suggested to be a risk factor for the development of mood disorders such as the burn out syndrome, anxiety, and depression. Experimental and clinical studies provide evidence that food intake restricted to the normal activity phase is a potent synchronizer for the circadian system and can prevent the detrimental health effects associated with circadian disruption. Here, we explored whether adult male Wistar rats exposed to an experimental model of shift‐work (W‐AL) developed depressive and/or anxiety‐like behaviors and whether this was associated with neuroinflammation in brain areas involved with mood regulation. We also tested whether time‐restricted feeding (TRF) to the active phase could ameliorate circadian disruption and therefore would prevent depressive and anxiety‐like behaviors as well as neuroinflammation. In male Wistar rats, W‐AL induced depressive‐like behavior characterized by hypoactivity and anhedonia and induced increased anxiety‐like behavior in the open field test. This was associated with increased number of glial fibrillary acidic protein and IBA‐1‐positive cells in the prefrontal cortex and basolateral amygdala. Moreover W‐AL caused morphological changes in the microglia in the CA3 area of the hippocampus indicating microglial activation. Importantly, TRF prevented behavioral changes and decreased neuroinflammation markers in the brain. Present results add up evidence about the importance that TRF in synchrony with the light–dark cycle can prevent neuroinflammation leading to healthy mood states in spite of circadian disruptive conditions.

    Induction of Core Circadian Clock Transcription Factor Bmal1 Enhances β Cell Function and Protects Against Obesity-Induced Glucose Intolerance

    Type 2 diabetes mellitus (T2DM) is characterized by β cell dysfunction due to impaired glucose-stimulated insulin secretion (GSIS). Studies show that β cell circadian clocks are important regulators of GSIS and glucose homeostasis. These observations raise the question whether enhancement of the circadian clock in β cells will confer protection against β cell dysfunction under diabetogenic conditions. To test this we employed an approach by first generating mice with β cell-specific inducible overexpression of Bmal1 (core circadian transcription factor; β-Bmal1OV). We subsequently examined the effects of β-Bmal1OV on the circadian clock, GSIS, islet transcriptome, and glucose metabolism in context of diet-induced obesity. We additionally tested the effects of circadian clock-enhancing small molecule Nobiletin on GSIS in mouse and human control and T2DM islets. We report that β-Bmal1OV mice display enhanced islet circadian clock amplitude, augmented in vivo and in vitro GSIS and are protected against obesity-induced glucose intolerance. These effects were associated with increased expression of purported BMAL1-target genes mediating insulin secretion, processing, and lipid metabolism. Furthermore, exposure of isolated islets to Nobiletin enhanced β cell secretory function in Bmal1-dependent manner. This work suggests therapeutic targeting of the circadian system as a potential strategy to counteract β cell failure under diabetogenic conditions.

    Elevated plasma branched-chain amino acid levels correlate with type 2 diabetes-related metabolic disturbances

    Total BCAA levels were higher in patients with T2DM compared to CON, but not in FDR, and correlated negatively with muscle mitochondrial oxidative capacity (r = -0.44, p<0.001). Consistently, whole-body leucine oxidation rate was lower in patients with T2DM vs. CON under basal conditions (0.202 ± 0.049 vs. 0.275 ± 0.043 μmol kg -1 min -1, p<0.05) and tended to be lower during high insulin infusion (0.326 ± 0.024 vs. 0.382 ± 0.013 μmol kg -1 min -1, p=0.075).

    In patients with T2DM, a compromised whole-body leucine oxidation rate supports our hypothesis that higher plasma BCAA levels may at least originate partly from a low mitochondrial oxidative capacity.

Murashige et al. measured more than 270 metabolites using liquid chromatography–mass spectrometry in human blood samples taken from an artery entering the heart and from a vein leaving it. Differences thus reflected the metabolic processes at work in the heart. Their results confirmed that hearts voraciously consume fatty acids. Hearts secreted, rather than consumed, amino acids, thus revealing active proteolysis. In patients with heart failure, ketone and lactate consumption increased, as did proteolysis. These findings could lead to strategies for fighting heart disease by altering metabolism

Jego zdaniem za ludzką świadomość odpowiada pole elektromagnetyczne. I stara on się swoją teorię poprzeć zmodyfikowaną wersją dualizmu ontologicznego.
    Jeszcze za czasów Kartezjusza, idea dualizmu ontologicznego według którego istnieje podział na ciało i duszę, była bardzo popularna. Z czasem jednak wielu badaczy uznało ją za mało naukową. Skierowali się ku teorii dotyczącej tego, że świadomość wytwarzana jest przez mózg i neurony w nim obecne. McFadden uważa jednak, że kluczowe jest tu rozróżnienie na nie "materię i ducha", a "materię i energię". Jaka to energia?

    Chodzi naturalnie o energię wytwarzaną przez mózg w formie pola elektromagnetycznego. Teoria badacza jest o tyle rewolucyjna, że pokazywałaby, czemu nie powstało jeszcze samoświadome AI, pozbawione w końcu zarówno "mózgu" jak i pola elektromagnetycznego.     Intermittent Fasting Improves Cardiometabolic Risk Factors and Alters Gut Microbiota in Metabolic Syndrome Patients

    Main Outcome Measure
    Cardiometabolic risk factors including body composition, oxidative stress, inflammatory cytokines, and endothelial function were assessed at baseline and 8 weeks. The diversity, composition, and functional pathways of the gut microbiota, as well as circulating gut-derived metabolites were also measured.

    39 patients with MS were included: 21 in the IF group and 18 in the control group. On fasting days, participants in the IF group reduced 69% of the calorie intake compared to nonfasting days. The 8-week IF significantly reduced fat mass, ameliorated oxidative stress, modulated inflammatory cytokines, and improved the vasodilatory parameters. Furthermore, IF induced significant changes in gut microbiota communities, increased the production of short-chain fatty acids (SCFAs), and decreased the circulating levels of lipopolysaccharides (LPS). Gut microbiota alteration attributed to the IF was significantly associated with cardiovascular risk factors and resulted in distinct genetic shifts of carbohydrate metabolism in the gut community.

    IF induces a significant alteration of the gut microbial community and functional pathways in a manner, which is closely associated with the mitigation of cardiometabolic risk factors. The study provides potential mechanistic insights into the prevention of adverse outcomes associated with MS.

    The Impact of Dietary Fiber on Gut Microbiota in Host Health and Disease

    Food is a primordial need for our survival and well-being. However, diet is not only essential to maintain human growth, reproduction, and health, but it also modulates and supports the symbiotic microbial communities that colonize the digestive tract—the gut microbiota. Type, quality, and origin of our food shape our gut microbes and affect their composition and function, impacting host-microbe interactions. In this review, we will focus on dietary fibers, which interact directly with gut microbes and lead to the production of key metabolites such as short-chain fatty acids, and discuss how dietary fiber impacts gut microbial ecology, host physiology, and health. Hippocrates’ notion “Let food be thy medicine and medicine be thy food” remains highly relevant millennia later, but requires consideration of how diet can be used for modulation of gut microbial ecology to promote health.

    Chronic n-3 fatty acid intake enhances insulin response to oral glucose and elevates GLP-1 in high-fat diet-fed obese mice

    n-3 polyunsaturated fatty acids (PUFA) can exert beneficial effects on glucose homeostasis, especially in obese rodents. Gut incretin hormones regulate glucose and lipid homeostasis, but their involvement in the above effects is not entirely clear. This study aims to assess the effects of chronic n-3 PUFA administration on the insulin and incretin responses in C57BL/6N obese male mice subjected to oral glucose tolerance test (oGTT) after 8 weeks of feeding a corn-oil-based high-fat diet (cHF). The weight gain and adiposity were partially reduced in mice fed cHF in which some of the corn oil was replaced with n-3 PUFA concentrate containing ∼60% DHA and EPA in a 3 : 1 ratio. In addition, these mice had improved glucose tolerance, which was consistent with an increased insulin response to oral glucose and plasma glucagon-like peptide-1 (GLP-1) levels. While the stimulatory effects of n-3 PUFA on GLP-1 levels could not be attributed to changes in intestinal or plasma dipeptidyl peptidase-4 activity, their beneficial effects on glucose tolerance were abolished when mice were pretreated with the GLP-1 receptor antagonist exendin 9–39. Moreover, chronic n-3 PUFA intake prevented the detrimental effects of cHF feeding on glucose-stimulated insulin secretion in the pancreatic islets. Collectively, our data suggest that n-3 PUFA may modulate postprandial glucose metabolism in obese mice through a GLP-1-based mechanism. The significance of these findings in terms of the effective DHA and EPA ratio of the n-3 PUFA concentrate as well as the effect of n-3 PUFA in humans requires further research.

    The Cancer Clock Is (Not) Ticking: Links between Circadian Rhythms and Cancer

    Circadian rhythms regulate many physiological and behavioral processes, including sleep, metabolism and cell division, which have a 24-h oscillation pattern. Rhythmicity is generated by a transcriptional-translational feedback loop in individual cells, which are synchronized by the central pacemaker in the brain and external cues. Epidemiological and clinical studies indicate that disruption of these rhythms can increase both tumorigenesis and cancer progression. Environmental changes (shift work, jet lag, exposure to light at night), mutations in circadian regulating genes, and changes to clock gene expression are recognized forms of disruption and are associated with cancer risk and/or cancer progression. Experimental data in animals and cell cultures further supports the role of the cellular circadian clock in coordinating cell division and DNA repair, and disrupted cellular clocks accelerate cancer cell growth. This review will summarize studies linking circadian disruption to cancer biology and explore how such disruptions may be further altered by common characteristics of tumors including hypoxia and acidosis. We will highlight how circadian rhythms might be exploited for cancer drug development, including how delivery of current chemotherapies may be enhanced using chronotherapy. Understanding the role of circadian rhythms in carcinogenesis and tumor progression will enable us to better understand causes of cancer and how to treat them.

    Emerging evidence demonstrates the importance of sufficient vitamin D (1α, 25-dihydroxyvitamin D3) levels during early life stage development with deficiencies associated with long-term effects into adulthood. While vitamin D has traditionally been associated with mineral ion homeostasis, accumulating evidence suggests non-calcemic roles for vitamin D including metabolic homeostasis. In this study, we examined the hypothesis that vitamin D deficiency (VDD) during early life stage development precedes metabolic disruption. Three dietary cohorts of zebrafish were placed on engineered diets including a standard laboratory control diet, a vitamin D null diet, and a vitamin D enriched diet. Zebrafish grown on a vitamin D null diet between 2–12 months post fertilization (mpf) exhibited diminished somatic growth and enhanced central adiposity associated with accumulation and enlargement of visceral and subcutaneous adipose depots indicative of both adipocyte hypertrophy and hyperplasia. VDD zebrafish exhibited elevated hepatic triglycerides, attenuated plasma free fatty acids and attenuated lipoprotein lipase activity consistent with hallmarks of dyslipidemia. VDD induced dysregulation of gene networks associated with growth hormone and insulin signaling, including induction of suppressor of cytokine signaling. These findings indicate that early developmental VDD impacts metabolic health by disrupting the balance between somatic growth and adipose accumulation.

    Effects of dietary macronutrients on liver fat content in adults: a systematic review and meta-analysis of randomized controlled trials

    Dietary macronutrient composition may affect hepatic liver content and its associated diseases, but the results from human intervention trials have been equivocal or underpowered. We aimed to assess the effects of dietary macronutrient composition on liver fat content by conducting a systematic review and meta-analysis of randomized controlled trials in adults. Four databases (PubMed, Embase, Web of Science, and COCHRANE Library) were systematically searched for trials with isocaloric diets evaluating the effect of dietary macronutrient composition (energy percentages of fat, carbohydrates, and protein, and their specific types) on liver fat content as assessed by magnetic resonance techniques, computed tomography or liver biopsy. Data on change in liver fat content were pooled by random or fixed-effects meta-analyses and expressed as standardized mean difference (SMD). We included 26 randomized controlled trials providing data for 32 comparisons on dietary macronutrient composition. Replacing dietary fat with carbohydrates did not result in changes in liver fat (12 comparisons, SMD 0.01 (95% CI −0.36; 0.37)). Unsaturated fat as compared with saturated fat reduced liver fat content (4 comparisons, SMD −0.80 (95% CI −1.09; −0.51)). Replacing carbohydrates with protein reduced liver fat content (5 comparisons, SMD −0.33 (95% CI −0.54; −0.12)). Our meta-analyses showed that replacing carbohydrates with total fat on liver fat content was not effective, while replacing carbohydrates with proteins and saturated fat with unsaturated fat was. More well-performed and well-described studies on the effect of types of carbohydrates and proteins on liver fat content are needed, especially studies comparing proteins with fats.

    słuchałem kilka ostatnich podcastów Kiefera na i porusza on tam te zagadnienia w ciekawym kontekście ale chciałbym uzupełnić swoją wiedzę jakimiś opracowaniami. Nie proszę Cię oczywiście o przeszukiwanie wątku ale jako główny kontrybutor może pamiętasz z głowy czy coś takiego wrzucałeś.
